Advances in CRISPR/CAS gene therapy in the approach to sickle cell disease:
a literature review

OBJECTIVES: The study aims to present and discuss the innovative applications of gene therapy, with a focus on the CRISPR/Cas9 technique, in the treatment of sickle cell anemia. METHODS: This is a systematic review of the literature in which the VHL and PubMed databases were used. RESULTS: Research has shown that gene editing with CRISPR/Cas9, especially Casgevy therapy, has shown promise in the treatment of sickle cell disease. The technique increases the production of fetal hemoglobin by inactivating the BCL11A gene or promoter regions of the HBG1/HBG2 genes. Clinical and preclinical trials indicate efficacy, safety, and therapeutic potential, although challenges such as graft efficiency and side effects still exist. CONCLUSION: Gene editing represents a promising therapeutic frontier for sickle cell disease, with the potential to transform the treatment of inherited diseases. Although the advances are significant, there are still challenges to overcome, which reinforces the importance of continuous research.
